Professional direction：The study revealed C-jun phosphorylation as a key regulator of complement activation in renal ischemia-reperfusion injury (Iri) through phosphoproteomics and functional validation. Iri is a common problem in kidney transplantation, affecting graft function. Treatment of the complement receptor CRIG/FH demonstrates the potential to improve IRI. The study found that CRIGHH treatment reversed the IRI-triggered Hyperphosphorylation state, and that differently expressed phosphoproteins were associated with cell adhesion and cytoskeleton reorganization. C-jun was identified as the most prominent phosphorylated transcription factor in complement activation. Immunoblotting confirmed the phosphorylation of C-jun and found that IRI upregulated the MRNA levels of TNF-Α, IL-6, Kim-1, and C-MYC, while CRIG/FH treatment restored these levels. JNK inhibitor decreased C-jun phosphorylation and alleviated C3D accumulation in tubular epithelial cells. Studies have shown that C-jun phosphorylation has an important role in IRI-related complement activation, providing new insights into the pathological mechanism of IRI.
